  11.  AB    To evaluate the performance of their diagnostic HIV-1 DNA PCR, 8 Belgian
  12.        AIDS Reference Laboratories established a multicentre quality control.
  13.        The centres tested a set of Belgian and African patient samples,
  14.        collected in Belgium, and the British Medical Research Council (MRC)
  15.        HIV-1 PCR reference reagent kit, containing plasmid HIV-1 DNA at several
  16.        dilutions in human carrier DNA with appropriate negative controls. No
  17.        false positive results were reported. All centres proved to be able to
  18.        detect 1 to 2 copies of HIV-1 DNA. Among the 17 Belgian and African
  19.        HIV-1 seropositives, some centres reported up to 4 indeterminate
  20.        results, mainly due to failure of the SK38-39, SK68-69 (Ou et al. 1988)
  21.        and/or gag881-882 (Simmonds et al. 1990) primers and to a bad performing
  22.        algorithm. Only the H1POL4235-4538 nested pol primer set, developed by
  23.        one of the centres, correctly identified the tested HIV-1 positive and
  24.        negative samples. To improve their performance, the centres decided to
  25.        evaluate these pol primers as a reference primer set and to standardize
  26.        the testing algorithm. On 10 additional Belgian and African patient
  27.        samples, all centres achieved a sensitivity and specificity of 100% when
  28.        adapting a standardized algorithm based on 3 HIV-1 primer sets, one of
  29.        which is the H1POL4235-4538 primer set.
